Just to reiterate what everyone else has said:

1. If you're not familiar with the base Pandemic Rules, play 2 or 3 games without stickers or the Legacy deck to get a feel for it before you start making permanent changes to your game.

1a. If you ARE familiar with basic Pandemic, READ THE RULES THOROUGHLY. The rules for Legacy are 95% the same, but the remaining 5% will trip you up if you just assume that you already know. Specifically, the initial board setup is different, and the way outbreaks are handled is slightly different.

2. When you hit a STOP card in the legacy deck, stop drawing, but read the rest of the back of the Stop card so you know when you'll need to start drawing cards again. Usually you'll draw cards just before a game and just after a game, but sometimes you're instructed to draw cards in the middle of a game and you don't want to miss those times. It's what the purple Reminder token is for.

3. Legacy cards NEVER tell you to open Packages (Package being the in game term for the sealed black boxes in your game). When a card tells you to open something, it ALWAYS refers to dossier doors (on the big red TOP SECRET dossiers you got). If you need to open a package, the dossier will tell you that. Speaking of which...

4. When you tear off a dossier panel, don't just look at the panel you removed. Also look at the space left behind. Sometimes it will be blank, but sometimes there will be more instructions there. (Mainly, instructions to open a package. See point 3.)

5. Some of the doors have new rules stickers that go in the rulebook. Before putting them in the book where they go, read them carefully, and then hand the sticker to another player and have them read it as well. It's amazing the things you miss while staring right at it. Another pair eyes always helps.

6. Don't try to second guess what things mean. When you're following instructions, do exactly what the instructions say. No more, no less. Sometimes it will seem like there's an obvious missing piece of text, only for the very next thing you open to correct the issue.

Very minor January spoiler: [o]How many people actually wrote C0dA on the board in marker, only to then immediately open the sticker that names it?[/o]

7. If (when) you do make a mistake, don't sweat it. Very few things will cause any permanent damage to the campaign as a whole. Just remember for the next time.

And, last but most certainly not least:

8. Have a great time!
